Perfect stereo separation is always going to be the best option, and impossible to do for most people for 8+ hours a day because it requires that the right ear hear only what's on the right channel and the left ear hear only what's on the left channel. This combined with the stereo-spatial audio effects is what forces the brain to kick into hemispheric synchronization in order to process the incoming data. To get this effect to kick in fully, you need headphones.
That's not to say you're not going to get great results through speakers, which at best can give you a high degree of stereophonic separation if you have them placed on either side of your head. But speakers will always project sound such that both ears are picking up both channels, to at least some degree.
If this were not the case, I would not suggest that people use speakers.
But... headphone will always give a better effect than speakers.
